Anastrozole is the generic name for the trade name drug Arimidex®. In some cases, health care professionals may use the trade name Arimidex® when referring to the generic drug name Anastrozole.

Anastrozole is used to treat early hormone receptor-positive breast cancer. It is also used for first-line treatment of hormone receptor-positive or hormone receptor-unknown advanced or metastatic (cancer that has spread) breast cancer. Anastrozole is also used to treat advanced breast cancer that has grown or spread after tamoxifen treatment. This medicine is used only in women who have already stopped menstruating (postmenopausal).

Many breast cancer tumors grow in response to estrogen. This medicine interferes with the production of estrogen in the body. As a result, the amount of estrogen that the tumor is exposed to is reduced, limiting the growth of the tumor.

 Anastrozole is in a class of medications called nonsteroidal aromatase inhibitors. It works by decreasing the amount of estrogen the body makes. This can slow or stop the growth of many types of breast cancer cells that need estrogen to grow.

The dose of Arimidex is one 1 mg tablet taken once a day. For patients with advanced breast cancer, the medication should be continued until tumor progression. Arimidex may not work as well if taken with tamoxifen or an estrogen medication.

Anastrozole comes as a tablet to take by mouth. It is usually taken once a day with or without food. Take anastrozole at around the same time every day. Follow the directions on your prescription label carefully, and ask your doctor or pharmacist to explain any part you do not understand. Take anastrozole exactly as directed. Do not take more or less of it or take it more often than prescribed by your doctor.

In this trial, between February 2004 and June 2010, 3,484 postmenopausal womenwith HR-positive early-stage breast cancer were randomized in 71 centers in Austria to receive either two years or five years of extended adjuvant therapy. All had undergone an initial five years of adjuvant endocrine treatment, either tamoxifen or other regimens containing aromatase inhibitors.

The trial’s primary endpoint was disease-free survival. Secondary endpoints included overall survival, contralateral breast cancer (cancer in the opposite breast), fractures, and toxicity.

Study Rundown: Metastatic hormone-receptor positive breast cancer has long been associated with poor survival outcomes due to resistance to traditional therapies over time. In a breakthrough study first reported in NEJM in 2012, researchers from the S0226 trial indicated that such patients may derive some benefit from a combination therapy including anastrozole (an aromatase inhibitor) and fulvestrant (a selective estrogen receptor degrader). The initial results followed patients for a period of three years and found that the combined therapy led to increased overall survival rates and disease-free progressive survival rates when compared to a group of patients taking anastrozole alone.  This current analysis expands on those initial results, including five additional years of follow-up data. Researchers found that their overall survival and disease progression-free survival rates were still higher among patients who received the combined therapy. They also found that the safety profiles of each arm were not significantly different. This study strengthens the case for the use of combined therapy among patients with metastatic hormone-positive breast cancer.
